Secondary assays
At the second stage of drug discovery, downstream the primary screening; it is desirable to evaluate confirmed hits in a secondary screening, providing more functional information.
The goal of these secondary cell-based assays is to represent natural function and physiological conditions.
Ideally, the generated cell lines should express the target genes and their downstream signals at natural levels.
To establish those assays, it is therefore crucial to enable the engineering of any labs cell line of interest, relevant for the tested drug.

Performing targeted transgene integration in a selected transcriptionally active genomic site is not only fast, it also has the advantage of integrating a single copy of each gene of interest (GOI) into the same chromosomal environment, resulting in comparable expression levels from one GOI to another and therefore enables high comparability. The use of stable and isogenic cell lines is a good way to achieve consistent and reliable data.
