Customize your lab’s own cell lines
An engineered meganuclease, co-transfected with the GOI (Gene Of Interest) carrying integration matrix, recognizes an endogenous genomic locus.
The meganuclease binds to the DNA and induces a double stranded break or ‘cut’. Natural repair mechanisms in the cell sense the damage and trigger homologous recombination to enable DNA repair. The co-transfected GOI, flanked by homology regions, is used as a template for the repair. The GOI is stably integrated or ‘pasted’ at the meganuclease recognition site.
cGPS® Custom Full Kits are specifically developed to achieve targeted integration in native cell lines. Whether you have derived proprietary clones, have introduced reporter assays, you can capitalize on your cell lines and apply these “plug-and-play” type of reagents.
Engineered meganucleases targeting an organism-specific endogenous gene can be applied to various cell lines derived from the same organism.
Our cGPS® Custom Full Kits contain the necessary ingredients to achieve powerful targeted integration in native cell lines (any cell line that bears a 24 bp sequence present in a chosen endogenous gene that is cleaved by an engineered meganuclease targeting this gene):
- Engineered Meganuclease: targets a 24 bp DNA sequence present in an endogenous gene,
- Integration Matrix: contains a Multiple Cloning Site (MCS) to clone your gene of interest. The MCS is flanked by other genetic elements that are part of the expression cassette (typically promoter and pA signal). The transgene expression cassette is flanked by left and right homology arms. The homology arms are homologous to the genomic regions flanking the meganuclease recognition site and drive homologous recombination and targeted transgene integration at the meganuclease recognition site.
