Targeted integration of shRNAs

Meganuclease-driven targeted integration can be used to generate stably silenced cell lines: any integrated shRNA will always be expressed from the same genomic position at the same level, bringing more robustness to the cellular assay.

Establishing stable and isogenic silenced cell lines is similar to the generation of overexpressing cell lines: instead of cloning a transgene expression cassette, the shRNA is cloned into an Integration Matrix adapted to shRNA expression.
