cGPS® Custom
Customize your lab’s own cell lines
cGPS® Custom TI kits are specifically developed to achieve targeted integration in native cell lines. Whether you have derived proprietary clones, have introduced reporter assays, you can capitalize on your cell lines and apply these “plug-and-play” type of reagents.
cGPS® Custom Targeted Integration Kits contain a engineered nuclease which target a native, endogenous genomic locus (e.g. a safe harbor). The engineered nuclease is co-transfected an Integration Matrix carrying the gene of interest (GOI). The nuclease recognizes specifically the endogenous genomic locus binds to the DNA and induces a double stranded break. Natural repair mechanisms in the cell sense the damage and trigger homologous recombination to enable DNA repair. The co-transfected GOI, flanked by homology regions, is used as a template for the repair. The GOI is stably integrated at the nuclease recognition site.
Our cGPS® Custom TI kits comprise all necessary reagents for targeted transgene integration in your own cell lines:
- Engineered nuclease: recognizes and cleaves a unique recognition site present at endogenous locus;
- Integration Matrix: a plasmid that contains a Multiple Cloning Site (MCS) to clone your gene of interest downstream of a promoter. The transgene expression cassette is flanked by left and right homology arms to drive homologous recombination for targeted transgene integration at the nuclease recognition site.
Engineered nucleases targeting an organism-specific endogenous gene can be applied to various cell lines derived from the same organism.

cGPS® Custom TI kits guarantee efficient selection of stable and isogenic clones with highly stable expression within 3 weeks, even in the absence of selection agents.

Our R&D team, composed of specialist in genome engineering, we developed a long list of optimized cell-specific protocols (transfection, nuclease expression, selection agents and concentration) for a range of human and rodent cell lines. We made available a wide range of TI protocol for the cell line of your choice.
Visit our dedicated section to select the cGPS® Custom protocol for your cell line of interest or contact us to discuss optimizing your own protocol.
Within the pIM store you can find a collection of Integration Matrices for a full-range of downstream applications, including gene expression regulation, gene tagging, integration of multiple expression cassettes at the same genomic locus, and many more.
