In drug discovery a first step is target identification. Primary screening (high throughput screening), conducted on millions of chemical or biological compounds, typically seeks to rapidly identify hits from compound libraries: which compounds bind to/inhibit targets of interest, to what degree of affinity.

A crucial issue is to establish reliable tools, as stable and robust cell-based assays, to carry out this screening with highest success to minimize the number of false positives and to maximize the number of confirmed hits.

Most cellular assays, based on transient expression system, show wide variability in gene expression, as a result of integration site and epigenetic regulation due to multi-copy integration and genomic instability.

Furthermore, these methods are time consuming and expensive. Using Cellectis bioresearch nuclease-driven targeted integration, stable and isogenic cell lines represent a good way to achieve consistent and reliable data.

Using targeted transgene integration in a selected transcriptionally active genomic site is not only fast, it also has the advantage of integrating a single copy of each gene of interest (GOI) into the same chromosomal environment, resulting in comparable expression levels from one GOI to another and therefore enables high comparability.

Cellectis bioresearch has developed cGPS® Targeted Integration (TI) kits for the fast and effortless generation of stable and isogenic cell lines from engineered cell lines.

Cellectis bioresearch also offers a Targeted Integration Service to generate stable and isogenic cell clones, or polyclonal population, expressing the customer's transgene of interest, by use of cGPS® TI kit.

Should you need to generate a specific stable cell line over expressing your gene of interest for HTS purposes, contact your local sales representative.
